The present study subsequently examined whether 5aand 5b induced the downregulation of cyclin D1 in either Sk-Hep1 orMDA-MB-231 cells, and where growth inhibition of Hep3B cells wascaused by cell cycle arrest. To address this question, the cellswere treated independently with 5a and 5b at ICconcentrations for 12 h, fixed, and cell cycle populations weredetermined using flow cytometry. The results demonstrated that thepercentage of the cell population in the G1 phase was significantlyhigher in all three cell lines, compared with the untreated controlgroup (). These resultsconfirmed that the growth of Hep3B cells was inhibited by cellcycle arrest, whereas the growth inhibition in Sk-Hep1 andMDA-MB-231 cells was mediated by Wnt/β-catenin signaling. Theexpression levels of cell cycle regulatory proteins, p21, p27 andCDK2, were also examined in the Hep3B cells. The expression levelsof p21 and p27 were significantly upregulated in the Hep3B cellsfollowing treatment with either of the two compounds, and CDK2 wassignificantly decreased following treatment (). In mammalian cells, cyclin D, Eand A are synthesized sequentially during the G1 phase of the cellcycle. The major catalytic partners of these cyclins are CDK2 andCDK4, which are negatively regulated by CDK-inhibitors, includingp21, p27 and p53 (,). The p53 tumor suppressor isrequired for the transcriptional activation of p21 (). Previously, indole-3-carbinoltetrameric derivative has been observed to induce G1 cell cyclearrest in breast cancer cells by upregulating p27kip1 (). In the present study, as expected,p21 and p27 were induced by 5a and 5b via a p53-independent pathwayin the Hep3B cells (p53 null type). These results suggested thatdownregulation of the expression of cyclin D1 and CDK2, and theinduction of p21 and p27, had a suppressive effect on the growth ofthe Sk-Hep1, Hep3B and MDA-MB-231 cells. Taken together, compounds5a and 5b inhibited the progression of cell cycle differentiallythrough Wnt/β-catenin mediation in the Sk-Hep1 and MDA-MB-231cells, and via p53-independent cell cycle arrest in Hep3Bcells.

This note describes a rapid and mild strategy for the loading of alcohols and anilines onto a polystyrene triphenylmethyl (trityl) resin. High loadings were obtained in a matter of minutes by treating resin-bound trityl chloride with triethyloxonium tetrafluoroborate followed by alcohols or anilines. Yields were comparable or better than known literature methods. Recycling of the recovered resin was also possible using the developed method.
