All nucleotides contain a ribose sugar and phosphate that form the backbone of DNA and RNA. These are synthesized from ribose 5-phosphate, a central metabolite of the pentose phosphate pathway. In this single step reaction, two of the phosphates of ATP are transferred to ribose 5-phosphate to form 5-phospho--D-ribosyl 1-pyrophosphate (PRPP). This intermediate is required for the biosynthesis of purines, pyrimidines, NAD, histidine and tryptophan. It plays a critical role in anabolism.

Nucleotides play a variety of important roles in all cells. They are the activated precursors of DNA and RNA. ATP, an adenine nucleotide, is a universal currency of energy in biological systems. GTP is an essential carrier of chemical energy. Adenine nucleotides are components of the coenzymes NAD+, NADP+, FMN, FAD and Coenzyme A. IMP is synthesized from ribose 5-phosphate. There are 11 reactions in the formation of IMP. Nucleoside monophosphates are converted to nucleoside diphosphates by base specific monophosphate kinases. Purine nucleotide synthesis is regulated by feedback inhibitor – AMP, GMP and IMP. Recycling of purines formed by the degradation of nucleotides is possible. Pyrimidine ring is synthesized as free pyrimidine and then it is incorporated into the nucleotide. Nucleotides of a cell undergo continuous turnover. Uric acid is the breakdown product of purine nucleotide. Gout is a disease characterized by elevated levels of uric acid in body fluids. Pyrimidines on degradation give rise to carbon dioxide, ammonia, β-alanine and β-amino isobutyrate.
Base pairing (French : appariement de bases) The pairing of nitrogenous bases in the polynucleotide chains by nitrogen bonds in a specific manner. The pairing occurs between a purine base of one strain and a pyrimidine of another strain in DNA, RNA or hybrid DNA/RNA molecules. In DNA the complementary bases pairs are Adenine and Thymine and Guanine and Cytosine. In RNA the base pairs are Adenine with Uracil and Guanine with Cytosine.

Biosynthesis of nucleotides is under tight regulatory control in the cell. Organisms need to make just the right amount of each base; if too much is made, energy is wasted, if too little, DNA replication and cellular metabolism come to a halt. Also, the cell is sensitive to the presence of any premade nucleotides in its environment and will down regulate their synthesis pathways in favor of using what is already present in the surroundings. Bacteria are capable of interconverting purines (adenine and guanine) and interconverting pyrimidines (thymidine, cytidine and uracil).
